Apologies if this has been asked before, I tried searching but could not find an answer.

Is it possible to tell xbmc to open a file and start playback at a specific time/position within that file? Can this be done from a plugin?

definitely from a plugin. just set the 'startoffset' property on the listitem prior to tossing it at the player. in seconds.
