I have a WIndows Vista PC running PlayOn. The PC serves content up to my XBOX 360, two DLNA compliant Blu-Ray players, and an HP MediaSmart TV (both DLNA and MediaCenter aware).
In this scenario, what will XBMC add to the equation?

A much better UI. I used to watch my shows via my Xbox 360/PlayStation 3, but navigating to find what I wanted to watch was tedious. XBMC has such a great UI that I enjoy browsing for shows. Heck, I'll sometimes leave it on on the main screen and just watch the backgrounds cycle.

OK, that answered the question for the XBOX360, what about other devices? What will it do for them?

It may not have answered the question for the 360 even. XBMC doesn't replace the UI of outside devices and doesn't have a DLNA server. Essentially, you'd be able to replace Windows Media Center. Beyond that, if you want the benefit of XBMC, you need to have a box with XBMC on it attached to each TV being used.
