I have a video called "video 1" and would like it to appear for two users.

If one users deletes it I do not want the video to be removed from the other user until both users have deleted it.

Is this possible in XBMC?

Do I just lookup XBMC Profiles and it automatically does the rest or do I need to have two copies of "video 1" one for Profile 1 and another for Profile 2?

You can have a separate library for each user using profiles (wiki), but there is "remove from library" and "delete". "Remove" will leave the file, but can be per user. "Delete" will delete the file totally.
