I have noticed that in at least one folder with a TV series which include subtitles, when I start an episode, the subtitle is always disabled, so each time I have to manually enable it. But on almost all all other shows/movies subtitles are enabled on start. Any idea on why or how to force subtitles to always be enabled?

It could be the settings of the subtitle stream in the video file. If there is no subtitle set as default and/or set to force, then it's possible.

Is it possible for me to force subtitles on in Kodi even if the stream says it's default off then?

Normally subtitles are enabled by default. Whichever gets preference is a Kodi program decision. I can only guess at this point that an additional 'Set to on' is necessary in case of a non-default or non-forced subtitle in the muxed video.
